Granzyme B is Critical for T Cell Receptor-Induced Cell Death of Type 2 Helper T Cells
Although CD95L is required for T cell receptor (TCR)induced cell death (TCR-ICD) in T helper 1 cells, the molecular mechanisms mediating TCR-ICD in Th2 cells are unknown. We found that death receptors were not involved in TCR-ICD of Th2 cells because blocking their cognate ligands had no effect on apoptosis of activated Th2 cells. Furthermore, we showed that caspases were not actively involved in TCR-ICD of Th2 cells. However, inhibition of granzyme B (GrB) activity abolished TCR-ICD in Th2 cells but not Th1 cells. Likewise, Th2 cells derived from GrB-deficient mice were resistant to TCR-ICD, and GrB deficiency or inhibition of GrB activity consequently enhanced the production of Th2 cytokines. GrB-deficient mice exhibited increased susceptibility to allergen-induced asthma. Thus, GrB plays a critical role in the TCR-ICD of Th2 cells. more...

"PIAS1 confers DNA-binding specificity on the Msx1 homeoprotein"
The mechanisms by which homeoproteins bind selectively to target genes in vivo have long remained
unresolved. Here we report that PIAS1 confers DNA-binding specificity on the Msx1 homeoprotein by
regulating its subnuclear localization and proximity to target genes. We demonstrate that the interaction of
Msx1 with PIAS1, but not its sumoylation, is required for Msx1 to function as an inhibitor of myoblast
differentiation through repression of myogenic regulatory genes, such as MyoD. We find that PIAS1 enables
Msx1 to bind selectively to a key regulatory element in MyoD, the CER, in myoblast cells and to distinguish
the CER from other nonregulatory TAAT-containing sequences. more...

Perspectives: NEUROSCIENCE: Enhanced: Neuron, Know Thy Neighbor
Our most sophisticated thoughts and feelings depend critically on the cellular composition and functional organization of the cerebral cortex. Indeed, changes in the numbers of neurons, their positions within the tissue, and their interconnections via synapses may underlie a variety of human brain disorders, from autism and bipolar depression to mental retardation and epilepsy. more...
